Scalable hiPSC Expansion on Microcarriers with BioThrust’s ComfyCell

High-density hiPSC expansion on microcarriers with lower shear and bubble-free oxygenation. In a 7-day perfusion process, BioThrust’s ComfyCell produced more than 1.5 billion hiPSCs, equivalent to a 65-fold expansion & while maintaining high viability, pluripotency-marker expression, and differentiation potential across all three germ layers.

Abstract

This study presents a benchmark evaluation of BioThrust’sComfyCell bioreactor for three-dimensional human inducedpluripotent stem cell (hiPSC) expansion on microcarriersunder dynamic conditions, demonstrating its suitability forefficient, high-quality, and scalable cell manufacturing. Usingthe ComfyCell, more than 1.5 × 10⁹ hiPSCs were produced,corresponding to a 65-fold expansion within 7 days, whilemaintaining high viability and preserving cellular identity.Differentiation assays further confirmed the potency ofharvested hiPSCs through successful generation ofderivatives from all three germ layers. In parallel, dissolvedoxygen and key metabolites were closely monitored,providing insights into culture conditions and enablingprocess optimization. Overall, these results demonstrate theComfyCell’s suitability for scalable hiPSC cultivation underdynamic conditions, supporting the development ofadvanced biomanufacturing platform solutions for stemcell–based therapeutics
