Simple hiPSC Expansion as Spheroids with BioThrust's ComfyCell Incu

Simple, incubator-compatible 3D hiPSC expansion with bubble-free membrane aeration. BioThrust’s ComfyCell Incu generated more than 800 million high-quality hiPSCs in a single 300 mL run while maintaining robust spheroid growth and greater than 99% core pluripotency-marker expression.

Abstract

This proof-of-concept study evaluates BioThrust’s ComfyCell Incu bioreactor for three-dimensional expansion of human induced pluripotent stem cells (hiPSCs) as spheroids. Using a simplified single-use 300 mL bioreactor format designed to support the transition from conventional 2D culture to 3D hiPSC expansion, the system produced up to 2.6×106 viable hiPSCs/mL and more than 8×108 cells in a single run, corresponding to a 17-fold expansion.

The ComfyCell Incu combines gentle hydrodynamics with diffusion-based gas transfer through hollow-fiber membranes, avoiding bubble sparging, reducing shear stress, and eliminating foam formation. hiPSCs formed robust aggregates, retained high viability and pluripotency, and showed more than 99% expression of core pluripotency markers, demonstrating a practical, incubator-compatible route toward scalable and reproducible 3D hiPSC manufacture.
